Route-16 Collection does not feature a continuous narrative in the traditional sense, as it is rooted in arcade-style gameplay. However, it presents a clear progression in concept and stakes across its three included titles.

In Route-16 and Route-16 Turbo, players are placed in a dangerous maze network where the objective is to collect scattered money while evading hostile vehicles and hazards. Survival and efficient navigation are the central themes, emphasizing risk versus reward as fuel runs low and enemies close in.

With Route-16R, the premise expands into a more action-driven scenario. Players pilot the advanced vehicle MAD X-R to reclaim the maze network from the Kanharoo Gang, a revived enemy force. Instead of merely escaping danger, players can now engage enemies directly by transforming into a combat robot, culminating in boss battles that must be cleared to progress.

Together, the collection reflects the evolution of the series—from simple arcade survival to an upgraded experience featuring combat, progression, and confrontation.
